Tremendous Advances in Transthyretin Amyloidosis Treatment: What Neurologists Need to Know

Transthyretin amyloidosis (ATTR) is a progressive disorder in which insoluble aggregates of amyloid fibrils deposit in various organs and tissues due to misfolded transthyretin protein. Accumulating amyloid fibrils leads to multisystemic dysfunction with mixed clinical phenotypes that include nonspecific autonomic, neurological, cardiovascular, and/or gastrointestinal symptoms. ATTR imposes a substantial burden on quality of life. In particular, its neurological symptoms, including peripheral and autonomic neuropathies (ATTR polyneuropathy, or ATTR-PN), are reported to cause the most substantial functional impairments, work limitations, and emotional distress.
